Thiocolchicoside is a muscle relaxant with anti-inflammatory and analgesic effects. It acts as a competitive GABAA receptor antagonist and also inhibits glycine receptors with similar potency and nicotinic acetylcholine receptors to a much lesser extent. It has powerful convulsant activity and should not be used in seizure-prone individuals.
Thiocolchicoside, a semi-synthetic derivative of the naturally occurring compound colchicoside extracted from the seeds of Gloriosa superba..Its Chemical name is N-[1,2-dimethoxy-10-methylsulfanyl-9-oxo-3-[(2S,3R,4S,5S,6R)-3,4,5-trihydroxy-6-(hydroxymethyl)oxan-2-yl]oxy-6,7-dihydro-5H-enzo[d]heptalen-7-yl] acetamide and molecular formula C27H33NO10S .

Thiocolchicoside is extracted from the seeds of Gloriosa Superba.
Gloriosa is a genus of five or six species in the plant family Colchicaceae, from tropical Africa and Asia. They are tender, tuberous rooted deciduous perennials, adapted to summer rainfall with a dormant dry season. Their native range is Africa, Southeastern Asia and parts of Malaysia, but they are now widely cultivated.
Gloriosa superba is the national flower of Zimbabwe, former Rhodesia (where it is a protected plant). It is also the state flower of Tamilnadu state in India.
Pharmacology
Thiocolchicoside binds to GABA-A and strychnine sensitive glycine receptors. Thiocolchicoside acting as a GABA-A receptor antagonist, its myorelaxant effects could be exerted at the supra-spinal level, via complex regulatory mechanisms, although a glycinergic mechanism of action cannot be excluded. The characteristics of the interaction of Thiocolchicoside with GABA-A receptors are qualitatively and quantitatively shared by its main circulating metabolite, the glucuronidated Derivative.
